Covid-19 antigen detection technology and solution

Corona virus disease 2019, referred to as Covid-19. At present, novel coronavirus pneumonia has been diagnosed in about 20,000,000 cases worldwide, which poses a great threat to global public health safety. In order to meet the requirements of Covid-19 rapid detection, the FDA issued the first emergency use authorization (EUA) Notice for Covid-19 antigen detection on May 9, 2020. Covid-19 antigen detection is a new detection method, which can quickly detect unique protein fragments on or in the virus by collecting samples from the mouth or nose with a swab.

The difficulty of developing Covid-19 antigen detection reagents lies in obtaining highly sensitive and specific antibodies. Antibody preparation, matching and screening are tedious and time-consuming. Significance of Covid-19 antigen detection

Covid-19 antigen detection can directly detect whether the human body contains Covid-19. The diagnosis is fast, accurate, and requires less equipment and personnel. Using double antibody sandwich method and using two antigen specific antibodies to identify and combine different epitopes of a target antigen, the probability of cross reaction can be greatly reduced, thus improving its specificity. Antigen detection is cheaper, easier to produce and faster to detect, and is suitable for large scale COVID-19 detection.

Principle of Covid-19 antigen detection

The Covid-19 antigen is detected by colloidal gold. The N protein of Covid-19 can act as immunogen and stimulate the plasma cells to produce specific antibodies after the virus infects the human body. According to the principle of double antibody sandwich ELISA, the sample is dropped on the sample pad and passed through the binding pad successively by liquid chromatography, Detection line (t line) and quality control line (C line) on NC membrane. The binding pad contains labeled antigen-specific antibody, which can bind to the antigen (viral protein) in the sample. When the liquid flow reaches the detection line (t line), the second antigen-specific antibody fixed on this line will bind to the antigen again, and the positive result will be presented. Quality control line (line C) coated with IgY antibody, which can be combined with the antibody in the sample pad to judge whether the chromatography process is smooth.

1. The protein concentration of each batch may be slightly different, and the actual batch shall prevail.

2. Recommended dosage: 800 ~ 1200 samples can be processed per mg of antibody (1mg of two paired antibodies each)

3. Large plates of 60 * 300mm size contain supporting diluent. More than 70 pieces can be cut for 4mm, and more than 90 pieces can be cut for 3mm
